![]() If your SAB is good, your sterile technique is good, and this is done in a room you can shut the door to and prevent anything from disturbing the plates this will not cause any contamination. Letting them cool together as a stack greatly reduces condensation when poured as close to the optimal temperature as possible. This is important: Once finished with all 20 plates let the plates cool, stacked and untaped, in the SAB for at least an hour before inoculation or taping for long term storage. You'll get into a nice rhythm after a few plates. However, this time stack the newly poured plate on top of the others. Starting with the next bottom plate, repeat the process. Replace the lid of the plate and set it off to the side. This is fine, just don't remove your hands from the SAB or wait more than a second or two. The agar will be just hot enough to tolerate touching so you may need to set it down after a few plates just to let your hand cool down for a second or two. Pour the agar into the plate, just covering the entire bottom of the plate. If you ever flip a lid completely upside down in an SAB, consider it no longer sterile. More than that and it's difficult to pour it all before it cools. Here's my technique that greatly cuts down on condensation (but does not eliminate it entirely): Wait too long and you get lumpy agar, pour too hot and you get tons of condensation. So essentially you have a very narrow window to pour properly. However, Agar begins to solidify too much under that and becomes completely solid at 80° F.
